Where the 36-hour claim is written
Lilly's US label starts most men on 10 mg tadalafil as needed, taken before anticipated sex. The same paragraph allows a rise to 20 mg or a drop to 5 mg. Once per day is the frequency cap for that as-needed path. The 36-hour sentence sits next to that start, not in the marketing brochure alone.
Two labelled time-point studies measured successful intercourse after a single dose. At 24 hours, mean per-patient success was 56% on 10 mg and 42% on placebo. At 36 hours it was 56% on 10 mg and 33% on placebo. Those are group rates. They do not book your Saturday.
A stopwatch study in the same label asked how often a successful erection arrived within 30 minutes. On 10 mg that figure was 38%, against 35% on placebo. On 20 mg it was 52%. Thirty minutes is early for this molecule. Median time to peak is 2 hours. The range runs from 30 minutes to 6 hours.
Half-life in healthy adults is 17.5 hours. Food does not change absorption. That pair of facts is why a Friday 10 mg tablet can still be useful on Saturday without a second swallow. It is also why a Saturday 10 mg on top of Friday is a dose increase, not a refresh.

Once-daily tadalafil for ED starts at 2.5 mg at about the same time each day. It may rise to 5 mg. You do not time it to sex. That is the point. After the first days the level is simply there.
I will not let a man take 10 mg 'most days' and call it daily therapy. Ten milligrams as-needed and 2.5 mg daily are different products on the label even when the INN is the same. Mixing them stacks exposure.
Daily 5 mg is also the labelled dose for BPH symptoms, and for ED plus BPH together. Ready tomorrow is not erect tomorrow
Priapism is the medical name for an erection that will not stand down. It is an emergency, not the 36-hour promise. If anything like that lasts past 4 hours, get seen. Do not wait for the 'weekend pill' story to finish itself.
What 10 mg actually does is quieter. It blocks PDE5, the enzyme that breaks down cGMP after nitric oxide has already started the erection chemistry. Between episodes of arousal the tablet is simply present. You are carrying a drug, not carrying an erection into breakfast.

Strong CYP3A4 inhibitors raise tadalafil levels. Ritonavir and ketoconazole are the names I ask for by the bottle, not from memory. Dose cuts belong in the clinic that already owns those drugs. A 10 mg tablet bought beside a protease inhibitor is not the start I described above.
Swallow the whole tablet. Do not split a 20 mg in half and call it a 10 mg start unless the tablet is scored and your prescriber said so. Most Cialis-shaped tablets are film-coated. Guessing the split guesses the dose.
